چکیده

Alpha-galactosidase is a glycoside hydrolase enzyme that hydrolyses the terminal alpha-galactosyl moieties from glycolipids and glycoproteins. Pseudomonas sp. MCCMB3 an alpha galactosidase producer was isolated from the rock soil of Kulasekaram, Kanyakumari district, South India. Totally 10 strains were isolated, when compared with other strains, predominant enzyme activity has been observed in the strain of Pseudomonas sp. MCCMB3. The maximum amount of enzyme was produced using nutrient broth during optimization. Environmental and nutritional parameters were optimized for the enhanced enzyme production. The enzyme production was maximum at pH 8.0 and temperature 50C. Glucose and ammonium carbonate induces enzyme production as carbon and nitrogen sources respectively. Based on the zymographic analysis, the molecular weight of the purified enzyme was about 80 KDa. The alpha galactosidase enzyme produced by Pseudomonas sp MCCMB3 degrades raffinose and stachyose present in soymilk which is used in other industrial applications.

β-galactosidase Production by Aspergillus niger ATCC 9142 Using Inexpensive Substrates in Solid-State Fermentation: Optimization by Orthogonal Arrays Design

Background: Enzymatic hydrolysis of lactose is one of the most important biotechnological processes in the food industry, which is accomplished by enzyme β-galactosidase (β-gal, β-D-galactoside galactohydrolase, EC 3.2.1.23), trivial called lactase. Orthogonal arrays design is an appropriate option for the optimization of biotechnological processes for the production of microbial...

Reduction of non-digestible oligosaccharides in soymilk: application of engineered lactic acid bacteria that produce alpha-galactosidase.

Human consumption of soy-derived products has been limited by the presence of non-digestible oligosaccharides (NDO), such as the alpha-galactooligosaccharides raffinose and stachyose. Most mammals, including man, lack pancreatic alpha-galactosidase (alpha-Gal), which is necessary for the hydrolysis of these sugars. However, such NDO can be fermented by gas-producing microorganisms present in th...

Biological Removal of phosphate from Synthetic Wastewater Using Bacterial Consortium

The biological phosphorus removal is a microbial process widely used for removing phosphorus fromwastewater to avoid eutrophication of water bodies. The study was aimed to screen the efficient phosphatereducing isolates and used to remove phosphate from synthetic wastewater using batch scale process. Thethree most efficient phosphate reducers were isolated and screened from eu...

Degradation of flatulence-causing oligosaccharides in soymilk by α-galactosidase—A novel thermotolerant from Penicillium purpurogenum

α-Galactosidase from Penicillium purpurogenum was induced to a greater extent in the culture medium by arabinose, galactose, lactose and guar galactomannan. The enzyme was purified by acetone precipitation and DEAE-Sephacel column chromatography. The purified enzyme showed a single band in both native-PAGE and SDS-PAGE. The molecular mass of enzyme was found to be between 107 and 110 kDa. The e...

Sequential degradation of keratan sulfate by bacterial enzymes and purification of a sulfatase in the enzymatic system.

Pseudomonas sp. IFO-13309 and Actinobacillus sp. IFO-13310, bacteria which exhibit a symbiotic growth in a medium containing keratin sulfate as a sole carbon source, were isolated from soil. Extracts of these organisms were shown to contain an endoglycosidase, a sulfatase, and exo-beta-D-galactosidase, and an exo-beta-D-N-acetylglucosaminidase which, together, catalyze an extensive cleavage of ...
